The present study aims to develop an efficient protocol for in vitro callus production, form Egyptian henbane plants using different explants using Murashige and Skoog medium supplemented with different concentrations and combinations of kinetin and 2,4-dichlorophenoxyacetic acid. Results showed that the callus formation depending not only on the concentration and combination of plant growth regulator but also on the type of used explant. The maximum frequency of callus formation was detected using root explants on a modified Murashige and Skoog medium supplemented with 0.5 mg / l of kinetin in combination with 0.5 mg / l of 2,4-dichlorophenoxyacetic acid after two months.
