EVALUATION OF THE EFFICIENCY OF CHROMATOGRAPHIC SEPARATION OF CELLULAR PROTEINS OF THE CAUSATIVE AGENT OF MELIOIDOSIS

This article evaluates the effectiveness of various methods for isolating and concentrating protein fractions of intracellular components of the melioidosis pathogen. The aim of the study was to compare the effectiveness of chromatographic methods for isolating individual protein fractions of intracellular components of the melioidosis pathogen using the antigenically complete B. pseudomallei 57576 strain as an example. Methods. The study involved a collection strain of the melioidosis pathogen, B. pseudomallei 57576, and fractions isolated from the original bacterial suspension. The B. strains were disinfected, and then the cellular components and virions were separated using anion exchange, hydrophobic interaction, and size-exclusion chromatography. The preparations were analyzed using polyacrylamide gel electrophoresis and ELISA. Results. Thirty fractions of the material containing various components of the original bacterial suspension were isolated and analyzed. Hydrophobic components of the bacterial mass demonstrated the highest activity in the ELISA method with specific immune sera from mice after hydrophobic chromatography with 5RPC sorbents (titer 1:400). Hydrophobic chromatography proved optimal for this task, as it allowed for the most efficient concentration and purification of fractions with immunological properties.
